چکیده

Haemopoietic stem cells give rise to all the different mature circulating blood cell types by a process of differentiation, proliferation and development. T h e pluripotent haemopoietic stem cells are required throughout the lifespan of an animal because mature cells, such as erythrocytes and neutrophils, are short lived. T h e defining characteristic of the stem cell population is therefore its continued pluripotentiality: it can both generate lineage-restricted progenitor cells which proliferate and develop into mature cells and self-renew to maintain the population of stem cells throughout the life of the organism. T h e structure and regulatory mechanisms that govern blood cell production have been the subject of intense research. It is hoped that, by

Growth and development of haemopoietic cells: a deterministic process?

The in vitro methods used to study haemopoiesis fall into two distinct categories. Short-term colony forming assays have identified a number of potent soluble factors capable of maintaining survival, proliferation and differentiation of haemopoietic cells but not their self-renewal. In contrast, in long-term bone marrow culture, extensive self-renewal occurs in the absence of exogenous factors ...

The distribution of granulocyte macrophage colony- stimulating factor (GM-CSF) in human long-term bone marrow cultures (HLTBMC) was examined using two monoclonal antibodies raised using purified recombi-

The role of growth factors in promoting the survival, proliferation and development of multipotent haemopoietic stem cells and their more developmentally restricted progeny has been unequivocally established using short-term in vitro clonogenic culture systems (Fauser and Messner, 1979; Johnson, 1984; Toogood et al., 1980). In other culture systems, however, haemopoiesis occurs in the absence o...

Up-regulation of the connexin43+ gap junction network in haemopoietic tissue before the growth of stem cells.

The early developmental stages of haemopoiesis are thought to be regulated by paracrine growth factors and by the haemopoietic environment. Are gap junctions involved here? Gap junctions are structures in cell membranes allowing the direct transfer of ions and small molecules between adjacent cells and are known to be involved in development. We have found that although connexin43 gap junctions...

Mesenchymal Stem Cells in Immune-Mediated Bone Marrow Failure Syndromes

Immune-mediated bone marrow failure syndromes (BMFS) are characterized by ineffective marrow haemopoiesis and subsequent peripheral cytopenias. Ineffective haemopoiesis is the result of a complex marrow deregulation including genetic, epigenetic, and immune-mediated alterations in haemopoietic stem/progenitor cells, as well as abnormal haemopoietic-to-stromal cell interactions, with abnormal re...
